What are the three media supplied in the Bioavailability assay kit: Human 18 used for?
Liver A and B Basal media are used for seeding and maintaining primary human hepatocytes in the Multi-chip Dual-organ plate. The Gut/Liver Basal media has been designed for primary Gut/Liver coculture for up to 72 hours to accomadate a drug-dosing window.
What is the shelf life of the Bioavailability assay kit: Human 18 and how do I store it?
The Bioavailability assay kit: Human 18 will arrive in three drop shipments.
1. CN Bio shipment contains Dual-organ plates (store at room temperature), Liver and Gut/Liver coculture basal media and supplements (store at +4°C, or -20°C as per kit instructions).
2. LifeNet Heatlth shipment contains primary human hepatocytes and should be stored in liquid nitrogen.
3. RepliGut® Planar Jejunum kit will be drop shipped last as this kit needs to be used within within a week of being received by the customer. Cells from Altis Biosystems to be stored in liquid nitrogen with the remaining kit components stored at +4°C.
Where can I find the Bioavailability assay kit: Human 18 protocol?
The protocol is supplied with the shipment of your PhysioMimix Bioavailability assay kit: Human 18 along with media preparation instructions, safety data sheets and other documents.
How are the cells within the Bioavailability assay kit: Human 18 validated for use?
Primary human hepatocyte donors are first validated for 3D tissue growth, health and metabolic activity following CN Bio’s standard validation process. Adequately performing human hepatocyte donors are then co-cultured with RepliGut®– Planar Jejunum*, following the Bioavailability assay kit: Human 18 SOP, to check that dual-organ (Gut/Liver) functionality is maintained.
*The RepliGut – Planar Jejunum kit is sourced from/ validated by Altis Biosystems.
How can you correlate data generated using the Bioavailability assay kit: Human 18 to the clinic?
Data generated using the Bioavailability assay kit: Human 18 can be used to predict human-relevant ADME parameters via your in-house in silico modeling.
