How do I order a DILI assay kit: Human 24?

Why are there two types of media supplied with the DILI assay kit?

The DILI assay kit: Human 24 is supplied with two Liver Basal Media (Basal A and B). Each needs to be combined with a supplement and bovine serum to support the generation of liver tissue:

  • Liver Seeding Media: made from Liver Basal A Media plus supplements, is required for cell seeding in Liver-12+ plates and the first 24 hours of culture.
  • Liver Maintenance Media: made from Liver Basal B Media plus supplements, is used for the remainder of the experiment, including drug dosing.

Sufficient Liver Basal B Media is provided to run an experiment using two Liver-12+ plates, for a 14-day cell culture period.

What is the shelf life of the DILI assay kit?

Individual elements of the kit have a different shelf life, and thus it is important to keep track of these when using the kit.

Where can I find the DILI assay kit protocol books?

The protocol is comprised of three documents:

  • List of Materials. This details all materials required to run a 14-day study using the kit (including any materials not supplied as part of the kit). A printed version is provided within the kit and a printable version will be emailed to you.
  • Liver Tissue Generation Protocol Book. This document lists all materials required for media preparation and the generation of 3D liver tissues. Plus a step-by-step protocol. This will be supplied by email and is downloadable via a QR code on the Quick Start Guide (included in the kit).
  • Drug Dosing Protocol Book. The third document lists all materials required for a 10-day drug dosing experiment and associated media preparation. Plus a step-by-step protocol for drug dosing. This will be supplied by email and is downloadable via a QR code on the Quick Start Guide (included in the kit).

How are the primary human cells in the DILI assay kit validated?

Firstly, primary human hepatocytes (PHH) are validated in isolation, following our standard validation process.

Once PHH donors have been validated, they are co-cultured with Kupffer cell donors, following the kit’s protocol, to ensure that the functionality of hepatocyte and Kupffer cells is maintained. In addition, validated co-cultures are tested for DILI response using the positive control included in the kit.

Once a donor combination has been validated, it is reserved for kit use.

Is donor info available for the primary human cells used in the DILI assay kit?

Yes, information is available for all donors upon request. Contact us for more information.

How many DILI assay kit boxes should I expect to receive, and how many components are within each box?

Customers will receive a minimum of five boxes per kit order. The kit will arrive in two shipments: CN Bio (three boxes) and LifeNet Health (two boxes) and contains storage condition information for the individual elements of the kit.

What is the standard experimental timeline for the DILI assay kit?

A standard experiment using the PhysioMimix DILI Assay kit: H24 takes up to two weeks and is usually split between two parts:

  • Tissue generation: primary liver tissue takes four days to stabilize. Quality control testing is carried out on day 4.
  • Drug dosing: occurs over the next ten days, depending on the toxicity of the tested compounds.

In addition to the DILI assay kit, what do I need to start my experiment?

Anyone purchasing the kit must have a PhysioMimix OOC System with two Single-organ drivers (MD6).

You will also need some common cell culture reagents including FBS (bovine serum) and Bovine serum albumin (BSA) to prepare the Liver Seeding and Maintenance Media. aPES filters are also required to filter media stock solutions during media preparation. We also strongly recommend using wide-bore 200 µL pipette tips for seeding primary liver cells to reduce damage and stress.

Standard cell culture equipment, such as pipettes, pipette tips, 15- and 50-mL Falcon tubes, PBS, Trypan blue and haemocytometer, is also required.

We recommend purchasing Glo-P450 test kits (Promega) to QC liver tissue for metabolic activity, plus testing for LDH and Albumin production to ensure that liver tissues are healthy and functional.

How many DILI assay kits can I use at the same time?

The PhysioMimix Single-organ System can run up to six plates simultaneously. Each kit contains two Multi-chip Liver-12 + plates, therefore you can use one, two or three kits simultaneously. To run at full capacity (72 wells /run), you will need one Controller, two docking stations and six Single-organ drivers. Customers with only one docking station and three drivers can only use one kit/run.
